Principle
The Geno Plus Genomic DNA Extraction Maxiprep System uses the silica-gel membrane technology for simple and fast isolation of genomic DNA without phenol/chloroform or ethanol precipitation. Sample homogenization is not necessary since tissues are directly lysed by Proteinase K. The unique buffer system is optimized to allow selective binding of genomic DNA to the silica-gel membrane. The simple centrifugation protocol completely removes contaminants such as proteins, divalent cations, and secondary metabolites. Pure genomic DNA is eluted in water or low-salt buffer, ready to use. Special protocols and guidelines are provided for verious samples, such as paraffin-embedded tissues, culture cells, blood, mouse tails, bacteria and yeast.
New buffer formulation, including a RBC lysis buffer in Geno Plus system, alloow up to 10ml of whole blood and broad range of samples with very high genomic DNA extraction efficiency, purified genomic DNA up to 200ug is free from contaminants and enzyme inhibitors, and typically has A260/A280 ratios between 1.7 and 1.9. The purified genomic DNA is sized up to 50-kb with predominant fragments of 30-kb.
